Data Processing With The Pros

Difficult High-resolution Triclinic Lysozyme

Very high resolution data can present a challenge for processing, as exemplified by this dataset from a crystal of triclinic hen egg-white lysozyme collected at 0.65 angstroms resolution. NOTE: This dataset is very large (>120 Gb), so only a small fraction (4.2 Gb) will be used for the demonstration (filename root: 'p1lyso_a.####.img'). Be sure to type in the 'rsync' commands accurately below to avoid downloading the whole dataset.

Useful Information

Data kindly provided by: Zbigniew Dauter
Sample: Hen egg-white lysozyme (triclinic)
PDB Code: 2VB1
Date data were collected: June 30, 2006
Beamline where data were collected: APS beamline 19ID
Image file format: The image files were collected on an ADSC Quantum 315 detector.

Four options to download the data:

    Option 1 (easiest):

Use 'rsync' (linux/UNIX command) to download only the first 3 (of 63!) tarred/compressed image bundles (command is in between quotes and should only be one line):

"rsync -azv anonymous@bl831.als.lbl.gov:pub/tarballs/\*2vb1_data_\[1,2,3\]_\* ."

and you will be prompted to enter 'lysozyme'.

    Option 2:

Use 'rsync' (linux/UNIX command) to download only the first 60 (of >1500!) images (command is in between quotes and should only be one line):

"rsync -azv anonymous@bl831.als.lbl.gov:pub/APS/19-ID/2vb1/p1lyso_a\*.img ."

and you will be prompted to enter 'lysozyme'.

    Option 3:

Use Wget to download only the 'p1lyso_a.####.img' images (try this if you have a Windows OS).

    Option 4:

Download either the compressed image file bundles (preferred) or the individual image files (a cumbersome last resort; you probably want the compressed file bundles instead, but if you're absolutely sure, remember to only download the 'p1lyso_a.####.img files!) using your web browser.
